Priming of Memory But Not Effector CD8 T Cells by a Killed Bacterial Vaccine

Supplemental Figure 1. HKLM primes p60 specific CD8 T cells in the absence of LLO. 2x106 L9.6 TCR transgenic splenocytes were labeled with CFSE and transferred into BALB/c Thy1.1 mice. One day later, mice were infected intravenously with 2,000 live L. monocytogenes, immunized with two consecutive daily doses of 109 HKLM derived from wild type bacteria or the avirulent L. monocytogenes strain DP-2161, which lacks the structural region of the LLO gene, or left uninfected. Splenocytes were harvested 5 days later. The dot plots show CFSE staining on the horizontal axis and Thy1.2 staining on the vertical axis for live lymphocytes gated for expression of CD8 and Thy 1.2. Each plot shows results for a single animal and is representative of 2 mice per experimental group.

Supplemental Figure 2. HKLM does not induce LM specific memory CD4+ T cells. 6 CB6 mice per group were immunized with PBS, two doses of 109HKLM or 104 live L. monocytogenes, and rechallenged 21 days later with 104 live L. monocytogenes. Intracellular cytokine staining for IFNname and TNF was performed on total spleen cells 5 days after infection. The mean number of cytokine producing CD4 T cells is plotted on the vertical axis and standard deviations are shown.
